Small RNA expression and miRNA modification dynamics in human oocytes and early embryos
Description
This collection of files contains small RNA-sequencing (sRNA-seq) data generated from human oocytes, zygotes, and pre-implantation embryos that were donated for research under an informed consent. Small non-coding RNAs (sRNAs) play important roles during the oocyte-to-embryo transition (OET), when the maternal phenotype is reprogrammed, and the embryo genome is gradually activated.
sRNA sequencing libraries (n=27) were prepared from GV (n=6), Metaphase I (MI; n=3), and Metaphase II (MII; n=3) oocytes, zygotes (two-pronuclear zygote, 2PN, n=2; three-pronuclear zygote, 3PN, n=3), and 4- (n=5) and 8-cell (n=5) embryos using a low-input compatible library preparation method. The libraries were collected, prepared, and sequenced in two batches (batch I and batch II). We used sRNAbench (Aparicio-Puerta et al. 2019; Rueda et al. 2015) to map and annotate sRNAs that have been previously identified and in-house R scripts for downstream analyses. The provided files are in fastq.gz format with one file per sample.
The dataset that supports these findings is accessible for scientific purposes. Due to patient consent and confidentiality agreements, data access will be evaluated by the Data Access Committee according to Finnish legislation. Data can be accessed internationally with valid Data Access Agreement, but data processing is restricted to Finland.
